JOB TITLE:   STAFF RESEARCH ASSOCIATE

JOB DUTIES: Contribute to the development of Arc’s Virtual Cell Initiative by generating single-cell CRISPR screening data in diverse cancer cell lines.  Clone high-quality sgRNA libraries for pooled CRISPR screens.   Execute 10x single cell genomics workflows, including sample preparation, library generation, and library QC.  Collaborate with teams across Arc to develop experimental workflows that support multiple aspects of the Virtual Cell Initiative, including model training and biological model development.  Design and execute cell line engineering campaigns in immortalized cells to enable pooled genetic perturbation experiments.  Analyze experimental data using software such as excel, Prism, FlowJo, and assist in the analysis and interpretation of large CRISPR screening datasets.  Document your experiments using ELN and project management software.  Present work to internal stakeholders at team and project meetings.

MINIMUM REQUIREMENTS:

Master’s degree in molecular biology, genetics, biotechnology, or related life science field,  or foreign equivalent, plus five years of experience in the biotechnology industry.
Experience must also include:
● 4 years of experience in a lab-based research and development position
● 4 years of experience working with mammalian cell culture, and cell line engineering using lentivirus and/or techniques for targeted genomic integration.
● 4 years of experience with pooled CRISPR screening workflows and cell selection, including pooled guide RNA library cloning, lentivirus packaging and titering, scaling cell cultures for high-throughput screening campaigns, multicolor flow cytometry, and cell sorting-based cell enrichment utilizing BD FACSAria or equivalent instrumentation.
● 4 years of experience with molecular biology techniques, including plasmid design, high-fidelity PCR and qPCR, advanced cloning methodologies such as Gibson and/or Golden Gate cloning, medium to large scale plasmid purification, and nucleic acid QC analysis.
● 4 years of experience in NGS workflows, which must include at least three (3) years of experience executing probe-based single-cell transcriptomics and fixed RNA profiling workflows (such as 10x Genomics Flex, or equivalent), high-throughput NGS library generation, and library QC utilizing TapeStation and Qubit.
● 3 years of experience maintaining experimental documentation in an Electronic Lab Notebook (ELN, such as Benchling or equivalent) and conducting scientific data analysis utilizing FlowJo, GraphPad Prism, Microsoft Excel, and Python for Next-Generation Sequencing (NGS) quality control (QC) metrics.

In the alternative, employer will accept a Bachelors degree in molecular biology, genetics, biotechnology, or a related life science field, or foreign equivalent, plus seven years of experience in the biotechnology industry. Experience must also include:

● 4 years of experience in a lab-based research and development position
